Role of Na§ § Exchange in the Control of Intracellular pH and Cell Membrane Conductances in Frog Skin Epithelium

چکیده
I N T R O D U C T I O N Changes in ion permeability at one epithelial cell membrane are often associated with modifications in ion t ransport at the opposing membrane ("cross-talk") (Schultz, 1981; Diamond, 1982). The nature of the cellular signal for "cross-talk" is not known, however, in a companion paper (Harvey et al., 1988) we have shown that sodium and potassium conductances in frog skin epithelium are very sensitive to slight variations in intracellular p H (pHi. This raises the interesting possibility that the mechanism(s) regulating p H i could also influence transepithelial ion transport. An understanding of how this epithelium maintains and regulates cell H § is, therefore, essential in interpreting changes in pHi and their electrical effects. Role of Na+/H+ exchange in the control of intracellular pH and cell membrane conductances in frog skin epithelium
Ion-sensitive microelectrodes and current-voltage analysis were used to study intracellular pH (pHi) regulation and its effects on ionic conductances in the isolated epithelium of frog skin. We show that pHi recovery after an acid load is dependent on the operation of an amiloride-sensitive Na+/H+ exchanger localized at the basolateral cell membranes. The antiporter is not quiescent at physiolo...
متن کاملIntracellular pH controls cell membrane Na+ and K+ conductances and transport in frog skin epithelium
We determined the effects of intracellular respiratory and metabolic acid or alkali loads, at constant or variable external pH, on the apical membrane Na+-specific conductance (ga) and basolateral membrane conductance (gb), principally due to K+, in the short-circuited isolated frog skin epithelium. Conductances were determined from the current-voltage relations of the amiloride-inhibitable cel...

AMPHIBIAN SKIN SERVES as an extra renal organ for the regulation of ion transport across the epithelium (2). Ion channels and transporters present in the epithelium of frog skin assist in the regulation of the same. These channels and transporters are necessary for the maintenance of intracellular ionic concentrations, osmotic concentration and cell integrity, and function (4). Figure 1A depict...
متن کاملpH Changes in Frog Rods upon Manipulation of Putative pH-regulating Transport Mechanisms
Rod intracellular pH (pHi) in the intact frog retina was measured fluorometrically with the dye 2',7'-bis(2-carboxyethyl)-5(and-6)-carboxyfluorescein under treatments chosen to affect putative pH-regulating transport mechanisms in the plasma membrane. The purpose was to relate possible pHi changes to previously reported effects on photoresponses. In nominally bicarbonate-free Ringer, applicatio...

In this study the role of glutathione (GSH) in sulfur mustard -induced oxidative stress and cytotoxicity, in human skin fibroblast cell line (HF2FF) was evaluated. Sulfur mustard-induced superoxide radical and hydrogen peroxide formation were evaluated by determination of superoxide dismutase and catalase activity in cell lysate. The cytotoxicity of sulfur mustard was estimated by lactate dehyd...
